Medulloblastoma Spatial Transcriptomics Reveals Tumor Microenvironment Heterogeneity With High-Density Progenitor Cell Regions Correlating With High-Risk Disease
Chien et al. (Emory / Children's Healthcare of Atlanta, with NYU Langone; JCO Precision Oncology 2026): spatial transcriptomics of 18 pediatric medulloblastoma samples obtained at diagnosis, including two matched diagnosis–relapse pairs, stratified into high- and standard-risk groups; dense regions of quiescent progenitor-like tumor cells were enriched in high-risk patients, and tumor-associated astrocytes, macrophages and dysregulated vascular endothelium increased after relapse; immunohistochemistry validation in the same cohort. Small cohort with only two relapse pairs; no external validation cohort is stated.
