A perturbation atlas of mouse neural organoids resolves macroglial reactivity and nominates drivers of degeneration
Preprint (not peer-reviewed): Dupré et al. (Camp lab, Roche Institute of Human Biology) map mouse neural organoids spanning multiple brain regions and the retina; benchmarked against in vivo references, they reach postnatal neuro-macroglial transcriptomic states within a month. A single-cell perturbation screen of 13 Müller-glia-specific pathways maps cytokine-driven degeneration-like stress programs in photoreceptors and reactive Müller-glia states (including an Oncostatin M-driven chaperone-associated state), and gp130–STAT3 signalling is nominated as a targetable driver. Mouse, non-tumour; small (13-pathway) screen.
